Mrs G?s lemon posset (?3.80) in contrast was as good as it gets. Light, sharp and creamy, it was elevated by the crispness of candied lemon peel and a stonking honey and oat biscuit.
What else is there to say about The Dogs except that the service was rapid and friendly, and that it's really excellent value? Oh yeah, it goes without saying that our meal was the dog?s proverbials.? BERLIN (AP) ? The number of Germans without jobs ticked up in August amid growing signs the European financial crisis is taking its toll on the region's largest economy, the Federal Labor Agency reported Thursday.
Some 29,000 more Germans were out of work in August over July, raising the jobless total to more than 2.9 million. When adjusted for seasonal factors, the number of jobless rose by 9,000.
Both the adjusted and unadjusted unemployment rates remained unchanged at 6.8 percent, the agency said.
Germany has been counting on domestic consumption to help offset weakening exports within the eurozone, where several countries are in recession.
But the new figures raise concerns about consumer spending, said Carsten Brzeski, an economist with ING in Brussels, who noted that the rise in jobless was the highest August increase since 1993.
"The strong labor market has been one of the main drivers of German growth in the first half of the year," he said in a research note. "Looking ahead, however, it is doubtful whether private consumption can really take over the baton as main growth driver for the German economy ? today's numbers provide further evidence that the labor market is gradually losing steam."
Economists have been warning that the debt crisis in the 17-country eurozone could eventually catch up with Germany. The country's economy has over the past few years done better than the currency union as a whole, which is struggling with a crisis over too much government debt and recessions in several countries.
Following two years of strong economic growth, the German economy grew by only 0.3 percent in the second quarter. Still, unemployment remains low, in sharp contrast with rates of more than 20 percent in Spain and Greece, which are both in recessions.
ScienceDaily (Aug. 30, 2012) ? When the parasite responsible for malaria infects human red blood cells, it launches a 48-hour remodeling of the host cells. During the first 24 hours of this cycle, a protein called RESA undertakes the first step of renovation: enhancing the stiffness of the cell membranes.
That increased rigidity impairs red blood cells' ability to travel through the blood vessels, especially at fever temperatures, according to a new study from researchers at MIT, the Institut Pasteur and the Korea Advanced Institute of Science and Technology (KAIST).
This marks the first time that scientists have identified the specific role of the RESA protein in modulating blood circulation in an environment that mimics physiological conditions, and could help researchers determine new strategies to combat malaria, which infects an estimated 300 to 500 million people worldwide each year.
The study -- coordinated by MIT's Ming Dao, a principal research scientist in the Department of Materials Science and Engineering (DMSE); Monica Diez Silva, a research scientist in DMSE; and YongKeun Park, an assistant professor of physics at KAIST -- appears Aug. 30 in Scientific Reports, an online journal of Nature.
Other MIT authors are Subra Suresh, former dean of the MIT School of Engineering and Vannevar Bush Professor of Engineering who is currently on leave from MIT serving as the director of the National Science Foundation; Jongyoon Han, an associate professor in the Department of Electrical Engineering and Computer Science (EECS); Sha Huang, an EECS graduate student; recent PhD recipient Hansen Bow; and the late Michael Feld, professor of physics.
This paper is the culmination of a collaboration between MIT and Institut Pasteur begun in 2004 when Suresh spent a sabbatical in Paris. The new results on dynamic microcirculation significantly extend earlier static experiments by the Suresh lab and the Pasteur team published in 2007, in which they found that RESA makes the red blood cell membrane stiffer than that of a healthy red blood cell by binding to the cell's internal scaffolding, called the cytoskeleton.
"The new work combines the latest advances in genetics, microfabrication, nanomechanics and computational modeling by bringing together interdisciplinary and international teams," says Suresh, the senior author of the paper.
Parasite invasion
When a mosquito carrying the malaria parasite, Plasmodium falciparum, bites a human, the parasite invades the liver, where it divides to produce tens of thousands of merozoites -- the form that can infect red blood cells. After several days or weeks, these merozoites are released into the bloodstream.
In the next stage of infection, the merozoites take up residence inside the red blood cells for about 48 hours, producing more copies of themselves. The RESA protein is expressed only during the first 24 hours, which is known as the "ring stage" due to the circular shape of the merozoites. During this stage, RESA "almost singlehandedly" alters the mechanical properties of red blood cell membranes, Dao says.
For the new study, the researchers created a microfluidic system at MIT that allowed them to study how RESA influences malaria-infected cells as they circulate. As cells flow through the microfluidic channels, they encounter narrow openings that are about the same width as the smallest capillaries (about three micrometers, or about 4 percent of the thickness of the human hair). The team found that malaria-infected cells passed through these channels much more slowly than healthy cells, due to the increased rigidity of the cells caused by the RESA protein.
The researchers also studied infected blood cells as they flowed through a microbead filtration system created at the Institut Pasteur. This system, which consists of densely packed tiny spheres, mimics the structure of the human spleen, which filters blood and removes old blood cells.
While RESA made the infected cells more rigid, they were still able to make it through the microbead system, suggesting that they would also slip through the spleen undetected. "In the ring stage the majority of the infected cells still go past, producing new parasites," Diez Silva says.
The researchers also found that RESA's effects on membrane stiffness and deformability are enhanced at fever temperatures: At these higher temperatures, RESA stabilizes the cell membrane so that it won't disintegrate. This is advantageous to the parasite because it helps the cell remain intact until the end of the 48-hour cycle, allowing it to produce many more copies of itself.
Mohandas Narla, vice president for research at the New York Blood Center, says the study is impressive and one of very few to look at the effects of temperature on malaria infection.
"Malaria is a very widespread global disease, so there is a lot of research being done on it, but most of the people who study it focus on biophysical measurements without paying attention to the fever part of the story," says Narla, who was not part of the research team.
Stopping the cycle
During the second 24 hours of the red blood cell infection, when RESA is no longer expressed, the parasite starts producing other proteins that make the cells even more rigid. They also churn out proteins that cause the cells to clump together and stick to the walls of blood vessels. At the end of the 48-hour cycle, the infected red blood cells burst and release new copies of the parasite, which target new cells and start the cycle over again.
Because they are so sticky, infected blood cells can't reach the spleen during this stage, so there is no chance for them to be filtered out. The new findings suggest that it might be beneficial to seek drugs that boost RESA expression during the first half of the cycle, when cells are still able to get into the spleen, the researchers say. Then, because the cells would be more rigid than typical infected blood cells, the spleen might be able to capture more of them.
The researchers are now further exploring how the spleen interacts with malaria-infected cells. The new microfluidic system used to mimic microcirculation in the small blood vessels is also being further studied as a novel diagnostic tool for detecting malaria and other diseases.
The research was funded by the Singapore-MIT Alliance for Research and Technology Center, the National Institutes of Health, the KAIST Institute for Optical Science and Technology, and the French Agence Nationale de la Recherche.

The field of space heritage preservation is gaining momentum, and a recently authored bill aims protect the Apollo 11?s Eagle lunar lander touchdown site and all the artifacts that astronauts?Neil Armstrong?and Buzz Aldrin left behind on the lunar surface.
A leading champion of the?moon landing site preservation?campaign is Beth O?Leary, an associate professor of anthropology in New Mexico State University?s College of Arts and Sciences. ?I had one sad thought that with his passing, people may consider the site on the moon more ?historic,?" she told SPACE.com.

There?s a total of 190 tons of cultural material on the moon, O'Leary said, and there are about 106 artifacts that are specific to the?Apollo 11?site. The effort under way is to preserve the significant technological objects that got Armstrong and Aldrin to the moon for that historic flight.
California and New Mexico have already listed Tranquility Base on their state historic registers. The next step would be the National Historic Landmark designation.

Westwood and O?Leary also have been working with the International Council on Monuments and Sites (ICOMOS), the World Heritage List?s advisory body, over the last three years to make the case for the Apollo sites to be added to this list.
?Sites on the list now include Chaco Canyon, the Pyramids at Giza, Stonehenge,? O?Leary said in a press statement, ?so there are many sites on the Earth that are recognized by this group of people. Putting the first lunar landing on the list means we recognize this is an achievement for humanity.?

?The passing of an American hero, Neil Armstrong, saddened the world," Westwood said. "It also brings to light the importance of preserving Apollo sites on the moon, because the human experience on the lunar surface was realized by only 12 humans ? a staggeringly slim percentage of humankind.? Westwood said that these brave astronauts are only with us for a brief period of time, and a generation from now, they will have passed on, no longer able to speak to their experiences and the historical significance of their achievement.
?By preserving the sites they left behind on?the moon, we are, in a sense, preserving not only the historical event of the first lunar landing, but the tangible memory of the Apollo astronauts,? Westwood said.
Legal gray area
?Our ideas on lunar preservation were presented by our ICOMOS ?caseworker? ? for lack of a better word ? in Helsinki a month ago,? O?Leary said. She and Westwood are working on getting the Tranquility Base Act in front of Congress to secure National Historic Landmark status; co-sponsors for Lundgren?s bill are still needed.
Objects at Tranquility Base are in a legal gray area. O?Leary said. By treaty, however, countries own the property they have placed on the moon, but no one can own the lunar surface.

If you're a student, you may have harbored the fantasy of learning lessons while you sleep. Who wouldn't want to stick on a pair of headphones, grab some shut-eye with a lesson about, say, Chinese history playing in his ears ? and wake up with newly acquired knowledge of the Ming Dynasty?
Sadly, it doesn't work. The history lesson either keeps you from going to sleep, or it doesn't ? in which case you don't learn it.
But researchers may have taken the first baby step to making the fantasy come true: In an unusual experiment published in Nature Neuroscience, researchers Anat Arzi, Ilana Hairston and others showed that people are capable of learning simple lessons while fast asleep.

The researchers sprayed volunteers with pleasant and unpleasant smells while they were fast asleep. Exactly like people who are awake, the sleeping volunteers took bigger breaths when there was a pleasant smell and a smaller breath when there was an unpleasant smell.
The researchers also paired each kind of smell with a particular sound. After repeated pairings of, say, a pleasant smell with a high-pitched beep, the researchers played the high-pitched beep without the smell: They found the volunteers took a big gulp of air. At least subconsciously, in other words, they were expecting the pleasant smell.
The researchers similarly paired an unpleasant smell ? rotting fish ? with another kind of tone. After repeated pairings, the tone alone caused the volunteers to take a smaller breath.
The phenomenon is known as conditioning, a simple form of learning made famous by Ivan Pavlov and his dog.
When the volunteers woke up, the researchers played the different tones to them. They found that the high-pitched beep caused the volunteers to take a big breath and the low-pitched beep caused the volunteers to take a small breath.
In other words, the conditioning lesson learned while the volunteers were asleep stayed with them after they awoke. Interestingly, the volunteers had no conscious recollection of having learned anything while they were sleeping.
Hairston said the research could help unlock the effects that sleep has on different brain processes. It's been known for a while that a nap can help cement things you've learned; Hairston said the sleep mechanisms that consolidate knowledge may explain why volunteers in the current experiment were able to bring what they learned during sleep into waking hours.
Practical applications of the conditioning technique are still far off, of course. But speculating about potential future uses, Hairston said: "A medical student can learn a list of body organs. You can perhaps pair different organs with smells. So you pair the thalamus with the smell of lemon, and the hypothalamus with the smell of orange blossom. When you wake up, you will have a better recollection of that list!"
ORLANDO, Fla. (AP) ? A three-judge panel in Florida has ruled that a former neighborhood watch leader charged in the fatal shooting of teenager Trayvon Martin should be granted a new judge in his case.
Florida's Fifth District Court of Appeal ruled Wednesday that Judge Kenneth Lester should enter a motion to disqualify himself in George Zimmerman's second-degree murder case. Zimmerman's attorney Mark O'Mara asked the court earlier this month to overturn a previous ruling by Lester not to leave the case.
One of the three judges dissented in the ruling.
O'Mara had argued that Lester should disqualify himself after he said the judge made disparaging remarks about Zimmerman's character and advocated for additional charges against him in setting his $1 million bond in July.
Zimmerman remains free on bail. He's pleaded not guilty.

Antibiotic residues in uncured pepperoni or salami meat are potent enough to weaken helpful bacteria that processors add to acidify the sausage to make it safe for consumption, according to a study to be published in mBio, the online open-access journal of the American Society for Microbiology, on August 28.
Sausage manufacturers commonly inoculate sausage meat with lactic-acid-producing bacteria in an effort to control the fermentation process so that the final product is acidic enough to kill pathogens that might have existed in the raw meat. By killing the bacteria that produce lactic acid, antibiotic residues can allow pathogenic bacteria to proliferate.
Researchers at the University of Copenhagen, Denmark, and University College Cork, Ireland, found that antibiotic concentrations within limits set by US and European Union (EU) regulators are high enough to slow fermentation, the process that acidifies the sausages and helps destroy foodborne pathogens like Salmonella or E. coli.
"At low concentrations and at regulatory levels set by authorities, we could see that the lactic acid bacteria are more susceptible to the antibiotics than the pathogens are," says Hanne Ingmer, of the University of Copenhagen, a researcher on the study. "So basically, we can have a situation where residual antibiotics in the meat can prevent or reduce fermentation by the lactic acid bacteria, but these concentrations do not effect survival or even multiplication of pathogens."
Antibiotics used as growth promoters or to treat disease in livestock can eventually end up in meat, and regulators in the US and EU have set limits on the concentrations of antibiotics in meat for consumption by humans. Ingmer and her colleagues set out to determine whether antibiotics falling within statutory limits might interfere with the process of fermentation in products like pepperoni, salami, or chorizo - sausages that are fermented using lactic- acid-producing bacteria in a curing process many cultures have employed for hundreds of years. She says fermented sausages occasionally cause serious bacterial infections, but it's never been understood why that might be.
In small-scale experiments in the lab, Ingmer and her colleagues added the antibiotics oxytetracycline or erythromycin to meat inoculated with lactic-acid-producing bacteria and pathogens Escherichia coli O157:H7 and Salmonella enterica. They followed the progress of the fermentation and tracked the survival of the pathogens. Ingmer says several different starter cultures of lactic-acid-producing bacteria were sensitive to these antibiotics and hence did not acidify the sausage meat effectively - results that could explain why people sometimes get sick from eating fermented sausage.
Ingmer says the results show antibiotics can potentially have a paradoxical effect that would increase the risk of foodborne illness: antibiotic residues reduce the effectiveness of bacteria that should make the sausages safe but don't affect the bacteria that can cause illness.
Although the results raise an alarm for the manufacture of processed meats, Ingmer stresses that it is important to conduct similar tests in manufacturing facilities. "The majority of sausages are manufactured at a commercial scale. It has to be addressed whether this is a problem in a real life facility," Ingmer says.
Manufacturers with good quality control systems can catch problems in fermentation, Ingmer says, preventing large-scale outbreaks of foodborne illness by testing the final product before it's distributed, but random batch testing could well miss a batch that didn't ferment properly, putting the public at risk.
How can we be sure antibiotic residues don't interfere with the safety of these products? Ingmer sees two possible solutions. If antibiotics are present in meat, boosting the survival and activity of the lactic-acid-producing bacteria is important. In the future, Ingmer hopes to work with manufacturers to develop cultures of lactic-acid-producing bacteria that tolerate low levels of antibiotics.
But the ultimate solution to the problem of antibiotics in meat may be harder to achieve. "The obvious solution is to eliminate the use of antibiotics as growth promoters and closely monitor the use of antibiotics in treating farm animal diseases," Ingmer says. The European Union and other countries have banned the use of antibiotics in livestock as growth promoters, Ingmer points out, a move the US is unlikely to follow very soon.

In the app, check-ins also are used to create a "Trending Tonight" list - a social hot list of the most popular TV shows.
Tanner views the user data as a game-changer for networks and ad agencies looking to predict the next big hit.
"We look at this as a potential early indicator of success," she said. Currently, the top show being added to watch lists is "Elementary" starring Jonny Lee Miller, airing this fall on CBS.
The app supports nearly every cable and satellite provider in the United States, although the streaming partners content is a bit more narrow and includes ABC, ABC Family, CW, Hulu Plus, HBO GO, MAX GO, Crackle and iTunes. Some popular streaming options - such as Netflix - are not yet supported.
"We have a couple dozen streaming partners in the queue so it's just a matter of time before we add more steaming partners in the app," Tanner said.
In addition, TV Guide plans to add more smart and social features such as remote record and recommendations.
The app is available in the United States, and select countries abroad in the Caribbean, South America and Europe.

In Making a Home Safer for an Elderly Family Member Part I, we discussed a number of objectives that should be achieved when executing a safety plan for a senior family member's home. This included a focus on interior and exterior stairs, outdoor walkways and high-traffic areas inside the home -- all of which can pose hazards if not situated and managed properly. In this second and final part of the series, we'll discuss the two rooms of most households that generally represent the highest level of danger in any home: the kitchen and the bathroom.
Bathroom hazards are often overlooked but in actuality are generally the most significant because use of this room is frequent and unavoidable. First, install no-slip pads under any rugs or mats. If possible, remove mats from in front of the sink and toilet, where tripping over an edge or having it slip out from under you is a serious concern.
Getting on and off the toilet requires strength and equilibrium; something we take for granted until our body is compromised in some way. To aide an elderly family member in this process, consider both a raised, padded toilet seat, and a safety bar attached to a nearby wall. In some cases installation of a phone or intercom system can aid in obtaining assistance in the case of trouble.
Safety bars are also essential in the bathtub or shower stall. Make sure the tub and shower floors have non-slip surfaces and invest in soap and shampoo racks that are designed to prevent the contents from sliding or falling out onto the floor. Towel racks can also inadvertently become "grab" bars, so it's a wise idea to see that they are well secured and can take the stress of being accidentally leaned on.
Check the temperature of the hot water as it comes out of the tap. If it scalds or is too hot to wash with comfortably at its highest level, turn down the temperature directly at the water heater, or have a professional manage this task for you.
Inventory the medicine cabinet. First-aid supplies -- bandages, anti-bacterial creams, etc, -- should be easily reachable. Outdated, long unused, or any medication that could be potentially harmful if accidentally ingested should be discarded. Over-the-counter headache or pain medications such as aspirin, ibuprofen or acetaminophen should be clearly marked and in bottles that can be opened easily.
Consider replacing shaving instruments with an electric razor. If an electric toothbrush or an electric razor is used, make sure that the electrical outlet is equipped with -- or replaced by one with --a ground fault interrupter or GFI receptacle -- a requirement in most modern homes.
Mouth rinsing glasses should be made of plastic, not glass -- as should any container holding grooming preparations.
A small bathroom mirror on an extendable rack can eliminate the need to lean over the vanity to peer into the wall mirror. Lighting in the bathroom is important must be adequate for the space, requiring replacement of insufficient bulbs with ones of higher output, and sometimes the installation of additional lights and fixtures.
Finally, be sure that both the path to the bathroom and the bathroom itself are equipped with wall socket nightlights so that nighttime trips to the bathroom are well marked and easily undertaken.
A local in-home care provider is often fully trained to spot hazards such as these and can quickly help you address them. This type of resource can prove invaluable, as many people overlook the most common danger areas -- even when actively seeking them out.
